Quantitative trait loci for resistance to Haemonchus contortus artificial challenge in Red Maasai and Dorper sheep of East Africa.

چکیده
A genome-wide scan was performed to detect quantitative trait loci (QTL) for resistance to the gastrointestinal nematode Haemonchus contortus in a double backcross population of Red Maasai and Dorper sheep. The mapping population comprised six sire families, with 1026 lambs in total. The lambs were artificially challenged with H. contortus at about 6.5 months of age, and nine phenotypes were measured: fecal egg count, packed cell volume decline, two weight traits and five worm traits. A subset of the population (342 lambs) was selectively genotyped for 172 microsatellite loci covering 25 of the 26 autosomes. QTL mapping was performed for models which assumed that the QTL alleles were either fixed or segregating within each breed, combined with models with only an additive QTL effect fitted or both additive and dominance QTL effects fitted. Overall, QTL significant at the 1% chromosome-wide level were identified for 22 combinations of trait and chromosome. Of particular interest are a region of chromosome 26 with putative QTL for all nine traits and a region of chromosome 2 with putative QTL for three traits. Favorable QTL alleles for disease resistance originated in both the Red Maasai and Dorper breeds, were not always fixed within breed and had significant dominance effects in some cases. We anticipate that this study, in combination with follow-up work and other relevant studies, will help elucidate the biology of disease resistance.

Exploring the Genetic Control of Resistance to Gastrointestinal Helminth Infections in Sheep and Mice
This paper reviews research being undertaken by ILRI and collaborators to identify Quantitative Trait Loci (QTL) for resistance to gastrointestinal (GI) nematodes in the resistant East African Red Maasai sheep and in inbred strains of mice. Six F1 rams, crosses of Red Maasai (resistant) and Dorper (susceptible) sheep, were bred and then backcrossed to pure Red Maasai and Dorper ewes to produce ...
متن کاملIdentification of quantitative trait loci affecting resistance to gastrointestinal parasites in a double backcross population of Red Maasai and Dorper sheep.
A genome-wide scan for quantitative trait loci (QTL) affecting gastrointestinal nematode resistance in sheep was completed using a double backcross population derived from Red Maasai and Dorper ewes bred to F(1) rams. This design provided an opportunity to map potentially unique genetic variation associated with a parasite-tolerant breed like Red Maasai, a breed developed to survive East Afric...
متن کاملDetection of quantitative trait loci for internal parasite resistance in sheep. I. Linkage analysis in a Romney x Merino sheep backcross population.
This study aimed to identify putative quantitative trait loci (QTL) that significantly affect internal parasite resistance in a backcross sheep population. A Romney x Merino backcross (to Merino) flock was challenged in 3 separate infections with Trichostrongylus colubriformis (primary and secondary) and Haemonchus contortus (tertiary). Haematological parameters were measured and faecal worm eg...
متن کاملIdentification of Novel Loci Associated with Gastrointestinal Parasite Resistance in a Red Maasai x Dorper Backcross Population
Gastrointestinal (GI) parasitic infection is the main health constraint for small ruminant production, causing loss of weight and/or death. Red Maasai sheep have adapted to a tropical environment where extreme parasite exposure is a constant, especially with highly pathogenic Haemonchus contortus. This breed has been reported to be resistant to gastrointestinal parasite infection, hence it is c...
متن کاملBreeding Programme and Infrastructure – the Case of Red Maasai Sheep in Kenya
The aim of this thesis was to study potential breeding strategies for indigenous livestock in Eastern and Southern Africa under low input production systems. The thesis covered a study of the status of supportive infrastructure for use of animal genetic resources. The case of Red Maasai sheep was studied as a model for design of strategies for improvement of an indigenous breed under threat. St...
